How to Read a Certificate of Analysis

A Certificate of Analysis, or COA, is the document that turns a purity claim into a verifiable fact. For anyone sourcing research peptides, it is the primary tool for confirming that the material in a vial matches what the label says. But a COA is only useful if you know how to read one. This walkthrough covers what a COA is, the three sections that carry most of the meaning, why the document has to be lot-specific, and what to ask when a supplier can't produce one.
What a COA actually is
A COA is a laboratory report describing the results of testing performed on a specific batch of material. At minimum, a meaningful COA identifies the compound tested, the lot it came from, the analytical methods used, and the results those methods produced. When the testing is done by an independent third-party laboratory, the COA also functions as outside verification — a record the supplier did not generate itself and cannot silently revise.
The point of a COA is not decoration. It exists so that a claim printed on a label can be traced back to measured data. A certificate that lists impressive numbers but omits the methods, the lot, or the testing party is closer to marketing than to analysis.
The identity section
The first thing a COA should establish is identity: is this compound actually the peptide named on the label? This is usually answered by mass spectrometry, which measures the molecular weight of the material and compares it against the known theoretical mass of the intended sequence. A COA reporting an observed mass that matches the expected mass is confirming that the molecule is what it claims to be — not a different peptide, not a shortened fragment, not a mislabeled product.
Identity matters because purity alone can't catch a substitution. A sample can be 99% pure and still be 99% the wrong compound. The identity section is what rules that out, and it is why a credible COA pairs a purity figure with a mass-spectrometry confirmation rather than reporting purity on its own.
The purity section
Purity is the number most buyers look for first, and it comes from high-performance liquid chromatography (HPLC). HPLC separates everything in the sample and measures how much of the total is the target peptide versus everything else. A purity of 99.5% means that 99.5% of the peptide content is the intended compound, with the remaining fraction made up of minor impurities — typically synthesis-related byproducts.
The most informative COAs show the chromatogram itself, not just the summary percentage. The chromatogram displays each component as a peak, so you can see the main peptide as a single dominant peak and judge whether impurities are negligible or significant. A clean chromatogram with one clear peak tells a fuller story than a lone number, and it is far harder to fake.
The net content section
The third section addresses a question that is easy to overlook: how much peptide is actually in the vial? Lyophilized peptides retain some residual water and counter-ions from manufacturing, so the gross weight of the powder is not the same as the mass of pure peptide. A vial sold as '10mg' is defined by its net peptide content — the actual milligrams of the target compound — not by total fill weight.
The net content figure on a COA tells you what the lot genuinely delivers per vial. It is the number that makes different suppliers' products comparable on a like-for-like basis, and its absence can quietly undermine a purity claim: a high percentage of a small or poorly quantified amount is not the same value it appears to be.
Why lot-specific documents matter
Peptides are produced in batches, and every section above describes properties of a particular batch. A COA that isn't tied to a lot code is describing a batch you may never have received. This is the difference between a certificate that proves something about your vial and one that proves something about some vial, once.
A trustworthy COA carries the lot number, and that lot number matches the code printed on the vial in your hand. Some suppliers make this connection effortless with a QR code on the label that links directly to the lot's certificate. However it is done, the link between the physical vial and the document is what gives the document its weight.
What to ask when a COA is missing
If a supplier can't produce a COA for the specific lot you're buying, a few direct questions cut through quickly. Was this lot tested by an independent third-party laboratory, and can you see that report? Does the certificate show both HPLC purity and mass-spectrometry identity data? Does the lot number on the document match the vial? What are the manufacture and expiry dates? And can you view the COA before purchase rather than after?
A supplier that answers these plainly, with documents rather than reassurances, is operating the way a research supplier should. One that deflects, offers only a generic certificate, or treats the request as unusual has answered the question in a different way. A COA is ultimately a test of transparency — and reading one well means knowing both what the document says and what its absence says just as loudly.
